Nutritional stress signaling through chromatin Our laboratory has extensive experience in the purification and analysis of protein complexes that modify chromatin for gene expression. This includes both affinity purification techniques and conventional chromatography. Examples of complexes first purified in this laboratory include the SAGA complex from yeast the Tip60 complex from Drosophila and the Sin3 complex from mammalian cells. We have extensively analyzed the mammalian Sin3 deacetylase complexes in collaboration with the Washburn laboratory. We have collaborated extensively with the…

TRIM24: an oncogenic histone reader in breast cancer Epigenetic factors are frequent targets of aberrant regulation, amplification or mutation in all human cancers. The interest in epigenetics for therapeutic development lies in the potential reversibility of epigenetic changes. Our laboratory focuses on Tripartite Motif Protein 24 (TRIM24), a multi-functional protein that i) negatively regulates p53 as an E3-ligase, ii) is a nuclear receptor co-regulator and iii) a PHD/bromodomain histone reader that recognizes a unique signature of histone PTMs (H3K4me0;H3K23ac). Recently,…

Epigenetics: A gatekeeper to DNA amplification and Drug response My laboratory studies how the chromatin microenvironment regulates gene expression while maintaining a stable genome.  We have initiated these types of studies by focusing on a specific class of chromatin regulators, JmjC-containing histone demethylases.  Since their discovery, my laboratory has been screening both solid and hematopoietic cancer for genomic anomalies in this class of enzymes.  We have uncovered the first enzyme (KDM4A- histone tri-demethylase) responsible for generating site-specific DNA copy gain…
