David Daniels MD PhD is a pediatric neurosurgeon and medicinal chemist at Mayo Clinic in Rochester MN. He is a clinician-scientist with a research focus in neuro-oncology and has a 50% research appointment. The majority of his clinical practice centers on treating children with brain tumors. David’s research interests revolve around the integration of neuro-oncology with neurosurgery. This includes developing new tools to improve surgical resection and decrease surgical morbidity for brain tumor surgery, and the development of novel drugs and delivery mechanisms for molecularly-targeted therapies. He completed neurosurgical residency at Mayo Clinic in 2013 and his pediatric neurosurgical fellowship at the University of Tennessee in Memphis/St Jude Children’s Hospital in 2014.
His lab is currently studying pediatric high grade brain tumors that harbor the histone H3K27M mutation to better understand how this mutation drives tumorigenesis. Relevant for their DIPG studies, they are developing patient-derived animal models and tumor cell lines with the H3K27M mutation to further study the mechanism of tumorigenesis and evaluate therapeutic vulnerabilities of this genetic mutation. They also collaborate with other labs that study the H3K27M mutation and have one of the largest collections of xenografts and cell lines with the H3K27M mutation. Utilizing this rich resource, they have performed extensive drug and shRNA screens and uncovered novel targets. Their laboratory is also evaluating alternative delivery mechanisms including CED, novel peptide carriers, and extracellular vesicles.
Dr. Daniels is very involved with education across the enterprise. He is the associate program director for the neurosurgery residency program at Mayo, he is on the MD/PhD admissions committee and on the Pharmacology admissions committee. He has mentored numerous MD/PhD and PhD students. In addition, he is associate medical director of pediatric trauma at Mayo Clinic.
